Current knowledge on bat lyssavirus infections in their native hosts is limited and little is known about the virulence, virus dissemination and transmission among free-living insectivorous bats. The present study is a brief description of rabies virus (RABV) dissemination in tissues of a naturally infected pregnant southern yellow bat (Lasiurus ega) and its fetuses, obtained by reverse-transcriptase polymerase chain reaction (RT-PCR). The RT-PCR was positive in samples from the brain, salivary gland, tongue, lungs, heart, kidneys and liver. Adaptation or acclimation to hypoxia occurs via the modulation of physiologically relevant genes, such as erythropoietin, transferrin, vascular endothelial growth factor, phosphofructokinase and lactate dehydrogenase A. In the present study, we have cloned, sequenced and examined the modulation of the LDH-A gene after an Amazonian fish species, Astronotus crassipinis (the Oscar), was exposed to hypoxia and anoxia. In earlier studies, we have discovered that adults of this species are extremely tolerant to hypoxia and anoxia, while the juveniles are less tolerant. Virus isolation and molecular characterization of canine distemper virus by RT-PCR from a mature dog with multifocal encephalomyelit BJM

A case of multifocal distemper encephalomyelitis in a mature dog is described. In the presented case the ante mortem clinical diagnosis of canine distemper virus (CDV) infection could not be ideally performed due to the absence of typical signs of distemper, such as myoclonus and systemic signs accompanying the nervous signs. The definitive diagnosis of distemper encephalomyelitis was only carried out at post mortem through virus isolation in cell culture from fresh central nervous system (CNS) fragments and CDV nucleoprotein gene detection in the CNS by RT-PCR.

Detección de potyvirus en plantas de pimiento (Capsicum annuum L.) y áfidos asociados al cultivo en Cuba Rev. Protección Veg.

El objetivo del trabajo fue detectar, mediante RT-PCR con cebadores genéricos, la presencia de potyvirus en plantas de pimiento (Capsicum annuum L.) con síntomas de moteado y en áfidos asociados al cultivo en los principales polos productivos de las regiones oriental, central y occidental de Cuba. Se extrajo el ARN total y se sintetizó el ADNc de las muestras colectadas. Se identificaron los áfidos como Myzus persicae (Sulzer). Se logró la detección de potyvirus en las muestras de las tres regiones, lo que demuestra que los potyvirus están ampliamente distribuidos en las áreas de producción del cultivo del pimiento en el país.

La anormalidad citogenética más común en la leucemia mieloide crónica (LMC) es el cromosoma Philadelphia, producida por la t(9;22), cuya expresión molecular es el gen de fusión BCR-ABL, que codifica proteínas con actividad tirosinquinasa. Según el punto de ruptura de los genes BCR o ABL se produce una proteína de fusión de 210-kD(p210) o 190-kD(p190). La presencia de este gen de fusión en pacientes con LMC tiene implicancia diagnóstica. Con el propósito de detectar transcriptos de fusión del gen BCR/ABL en pacientes con leucemia mieloide crónica, procedentes de la IX Región de Chile, se estudiaron 14 muestras de sangre de 11 pacientes con LMC. Detección cuantitativa del virus psorosis de cítricos mediante RT-PCR tiempo real Agrociencia

La RT-PCR tiempo real es útil para detectar el virus psorosis de cítricos (CPsV) y además permite evaluar la concentración viral que se puede usar para estudiar algunos aspectos de la enfermedad. En este trabajo se implementó un protocolo de RT-PCR tiempo real para la detección cuantitativa del CPsV, por lo que se diseñó un conjunto de iniciadores y una sonda Taqman con fundamento en una secuencia de 559 bases del gen responsable de la síntesis de la proteína de la cápside del CPsV procedente de tejido infectado de un árbol de campo. Vinte e nove pintos SPF de um dia foram inoculados com o vírus da doença infecciosa da bursa de Fabricius (VDIB) para avaliar a ocorrência precoce de apoptose e a expressão da proteína viral 2 (VP2) e da enzima gliceraldeído fosfato dehidrogenase (GAPDH). Determinou-se a expressão gênica das caspases 3 e 8 mediante transcrição reversa de mRNA total e reação em cadeia da polimerase (RT-PCR) para avaliar a apoptose em timo e baço de ratas imunossuprimidas por glicocorticóides. Utilizou-se dexametasona para indução da apoptose e atrofia linfóide. Human respiratory syncytial virus detection in children admitted at a community hospital in Botucatu, SP, Brazil BJM

Human Respiratory Syncytial Virus (HRSV) is recognized as the single most important pathogen responsible for acute viral infection of the lower respiratory tract in young children. In this study, 84 samples from children under two years of age presenting acute respiratory symptoms were obtained from September 2000 to November 2001. Analysis by immunofluorescence and reverse-transcription followed by PCR, revealed that 18% (15/84) of all the samples were positive for HRSV, in which 80% (12/15) of the cases was observed in children under six months of age and the subgroups A and B co-circulated in the studied period. These are the first data obtained for Botucatu city and seasonality is evident by the higher circulation of the virus between May and July.

Sm14 is a 14-kDa vaccine candidate antigen from Schistosoma mansoni that seems to be involved in cytoplasmic trafficking of fatty acids. Although schistosomes have a high requirement for lipids, they are not able to synthesize fatty acids and sterols de novo. Thus, they must acquire host lipids. In order to determine whether Sm14 is present in different stages of the life cycle of the parasite, we performed RT-PCR. Molecular analysis of the N gene of canine distemper virus in dogs in Brazil Arq. Bras. Med. Vet. Zootec.

Eleven central-nervous-system samples collected from stray dogs between 2000 and 2004 were found positive by RT-PCR, which amplified a 480bp fragment of the N gene of canine distemper virus (CDV). Phylogenetic analysis based on partial N-gene sequences showed four major clusters. All dog strains segregated into cluster I, with a mean nucleotide identity of 95.8% and 95.6% with the Onderstepoort and Lederle vaccine strains, respectively. Cluster II contained all the raccoon-related strains, cluster III Orient strains and Cluster IV the Onderstepoort and Lederle vaccine strains, with a mean nucleotide identity of 99.7% between them. This is the first report of phylogenetic analysis of CDV strains in Brazil.

A reverse transcriptase polymerase chain reaction (RT-PCR) to detect mouse hepatitis virus (MHV) in hepatic tissue was developed. To circumvent possible failures in RT-PCR amplifications, a second round of PCR with internal primers was used to confirm the specificity and increase the sensitivity of the test. Using this method specific amplification of MHV sequences was observed in 18 out of 20 mouse colonies examined.

The aim of the present study was to confirm the relevance of studying DNA adduct formation in a field study. In that context, freshwater mussels Dreissena polymorpha, collected in a reference station, were transplanted in different sites with a pollution gradient. After one and two months, mussels were collected and DNA adduct formation was analyzed using the 32P post labelling technique on both gills and digestive glands. Acute promyelocytic leukemia: the study of t(15;17) translocation by fluorescent in situ hybridization, reverse transcriptase-polymerase chain reaction and cytogenetic techniques BJMBR

Acute promyelocytic leukemia (AML M3) is a well-defined subtype of leukemia with specific and peculiar characteristics. Immediate identification of t(15;17) or the PML/RARA gene rearrangement is fundamental for treatment. The objective of the present study was to compare fluorescent in situ hybridization (FISH), reverse transcriptase-polymerase chain reaction (RT-PCR) and karyotyping in 18 samples (12 at diagnosis and 6 after treatment) from 13 AML M3 patients. Bone marrow samples were submitted to karyotype G-banding, FISH and RT-PCR. At diagnosis, cytogenetics was successful in 10 of 12 samples, 8 with t(15;17) and 2 without.
